摘要: CXCR4 is the receptor for α-chemokine stromal cell-derived factor 1 (SDF-1) and has been shown to be expressed on a diversity of leukocytes. In this report, expression in cells megakaryocytic lineage role SDF-1 megakaryocytopoiesis were investigated. Using flow cytometry combination with reverse transcriptase-polymerase chain reaction (RT-PCR), we observed that bone marrow CD34+, CD61+ cells, blood platelets, leukemia cell lines all receptor. To examine megakaryocyte progenitors (colony-forming units-megakaryocyte [CFU-Meg]), CXCR4-positive -negative CD34+ populations separated from cultured plasma clot culture system. A subpopulation CFU-Meg was found fraction. The functional significance examined by studying effects SDF-1α migration proliferation progenitor vitro. We potently induced significantly enhanced adhesion mature megakaryocytes endothelium. No marked alone or thrombopoietin stem factor/kit ligand production vitro noted. These results demonstrate first time platelets its may modulate several aspects megakaryocytopoiesis. © 1998 American Society Hematology.
