Effects of Allopurinol on Endothelial Dysfunction: A Meta-Analysis

摘要: Objective: Several studies have assessed the effect of allopurinol on endothelial function, but these were relatively small in size and used different methods evaluating function. We conducted a meta-analysis to investigate both endothelial-dependent -independent vasodilatation. Methods: Electronic databases, Medline, PubMed, EMBASE, SCOPUS, EBSCO Cochrane Library Central Register Clinical Trials searched from January 1985 July 2013 clinical trials (randomized non-randomized) which sensitivity analysis assess contribution each study pooled treatment by excluding one at time recalculating for remaining studies. Treatment was significant if p 2 statistic (with substantial heterogeneity defined as values >50%). Results: The final consisted 11 (2 observational 9 randomized). For vasodilatation there 6 studies, including 257 patients, that evaluated flow-mediated dilatation 5 with 87 patients reported data forearm blood flow response acetylcholine or flow-dependent Overall, increase endothelium-dependent (MD 2.69%, 95% CI 2.49, 2.89%, = 319.1, I2 96%, 9.0, 44%, 0.11) showed no endothelium-independent Conclusions: found hyperuricemia is associated an improvement endothelial-dependent, not endothelial-independent
