Species and strain comparisons of immunosuppression by 2-methoxyethanol and 2-methoxyacetic acid.

摘要: 2-Methoxyethanol (ME) and its principal metabolite 2-methoxyacetic acid (MAA) have been shown in our laboratory to be immunosuppressive male Fischer 344 rats. In this study several strains of 12-week-old female rats mice were used compare the activity equimolar concentrations ME MAA on trinitrophenyl-lipopolysaccharide (TNP-LPS) antibody plaque-forming cell (PFC) response, which we previously demonstrated a sensitive end point. Female inbred Lewis, Wistar/Furth, outbred Sprague-Dawley dosed by gavage with either or at dosages 0.33 2.64 mmol/kg/day for 10 consecutive days. C3H C57BL/6J, hybrid B6C3F1, CD-1 similarly 0.66 5.28 MAA. All animals immunized day 9 dosing PFC responses evaluated 3 days later. Suppression response was observed all Lewis Wistar/Furth found most suppression levels as low While resulted TNP rat tested, such treatment did not suppress any mouse examined. These results indicate that under conditions rats, but mice, are immunosuppressed exposure, susceptibility immunosuppression differs among strains.
