Pure albumin is a potent trigger of calcium signalling and proliferation in microglia but not macrophages or astrocytes

摘要: Microglial activation is implicated in the neurotoxicity of neurodegenerative diseases. Raised intracerebral levels albumin are associated with pathology Alzheimer's disease, multiple sclerosis, and stroke where blood-brain barrier damage evident. We report here that treatment primary cultured microglia N9 microglial cell line pure albumin, or which fatty acids immunoglobulins remain attached (fraction V), induced a rise intracellular calcium. This calcium was mediated via Src tyrosine kinase phospholipase C. The albumin-induced response coupled to proliferation, prevented by BAPTA, U73122 PP2 but not mimicked thapsigargin. In contrast, peritoneal macrophages were resistant albumin- fraction V-induced responses whilst astrocytes TSA-3 astrocyte responsive V albumin.
