摘要: Peritoneal macrophages (M phi) from mice become cytotoxic after incubation in lymphokine (LK)-rich supernatants of antigen-stimulated spleen cell cultures. Tumoricidal activity is evident with M phi treated LK for 4 hr, becomes maximal 8-12 hr and decreases to control levels by 24-36 hr. To gain insight into LK-induced functional changes, the lipid composition cultured 0-36 was analyzed high pressure liquid chromatography. induced marked changes composition: cellular content cholesterol (CHOL) polyunsaturated fatty acids increased 2- 3-fold 8 when cells showed tumoricidal activity. Cellular acid returned 24 had lost These were not observed equal numbers supernatants. analyze further role CHOL unsaturated tumor cytotoxicity, enriched or linolenic (18:3) tested their ability kill 1023 cells. Within 1 culture, a 3- 4-fold increase 18:3 content. 18:3-enriched markedly tumoricidal, whereas controls delipidized medium alone saturated cytotoxic. CHOL-enriched tumoricidal; indeed, these inhibited killing treatment compared alone. results suggest that UFA aids, negates, expression cytotoxicity.
