摘要: We determined the potential for induced pluripotent stem (iPS) cells to differentiate into nucleus pulposus (NP)-like in mice. iPS were generated from tail-tip fibroblasts. used a pellet culture model with aim of determining applicability cell-based therapy intervertebral disc degeneration (IVD). The cell was cultured an NP basal medium comprising Dulbecco's modified Eagle's supplemented transforming growth factor beta 1, dexamethasone, ascorbate-2-phosphate, and 1% ITS-Premix. evaluated by quantitative reverse transcription polymerase chain reaction, immunohistochemical staining, biochemical composition. differentiation demonstrated protein mRNA expression levels proteoglycan, collagen II, aggrecan, CD24. Furthermore, increased hydroxyproline content dimethylmethylene blue staining that II glycosaminoglycan time. have shown mouse can be cells. Such proof-of-concept opens up possibility producing patient-specific relatively simple straightforward manner high efficiency. are confident such could immediately useful study IVD disease.
