摘要: Peripheral benzodiazepine receptor (PBR), a mitochondrial protein involved in cell proliferation and differentiation, carbonic anhydrase IX (CA IX), an intrinsic marker of hypoxia, have been studied the panel human breast (MCF-7, BT- 20, MDA-MB-453, MDA-MB-231) ovarian (A2780, A2780/CP, A2780/ADR, CH1, SKOV-3) carcinoma lines that differ by malignant progression. The expression both antigens was detected staining with PBR-specific 8D7 CA IX-specific M75 monoclonal antibodies quantitated flow cytometry. PBR related to mass density. Breast showed higher relative fluorescence intensity than lines, exception A2780/CP cisplatin-resistant subline comparable highly invasive MDA-MB-231 line. Among increased their potential. greater variability intensities did not correlate amount mitochondria. Mitochondrial density disclosed significant difference between cisplatin-sensitive (low density) -resistant (high lines. MTT test sensitivity 2 MCF-7 (IC50 90 microM, chemo- radio- resistant IC50 > 110 microM). Growth inhibitory effect PK 11195 mediated probably another, PBRindependent mechanisms. only marginal majority tested subconfluent conditions inducible high More 5% positive cells sparse culture found MDA-MB-453 while more 15% A2780/ADR adriamycin-resistant were for under same conditions. Our data indicate is proportional mitochondria should be expressed relatively mass. This assessment allows establishing as measure aggressiveness (invasion resistance cancer). Observation evokes assumption multidrug might connected selection advantage towards expressing cells.
