Immunotherapeutic Potential of Mollusk Hemocyanins in Combination with Human Vaccine Adjuvants in Murine Models of Oral Cancer.

摘要: Mollusk hemocyanins have been used for decades in immunological and clinical applications as natural, nontoxic, nonpathogenic, nonspecific immunostimulants the treatment of superficial bladder cancer, carriers/adjuvants tumor-associated antigens cancer vaccine development adjuvants to dendritic cell-based immunotherapy, because these glycoproteins induce a bias towards Th1 immunity. Here, we analyzed preclinical therapeutic potential traditional keyhole limpet hemocyanin (KLH) two new from Concholepas concholepas (CCH) Fissurella latimarginata (FLH) mouse models oral squamous cell carcinoma. Due aggressiveness deadly malignant this were applied combination with adjuvants, such alum, AddaVax, QS-21, which shown be safe effective human vaccines, potentiate their antitumor activity. The immunogenic performance was compared, best formulation evaluated its effects murine cancer: MOC7 cells implanted flank (heterotopic) bioluminescent AT-84 E7 Luc floor mouth (orthotopic). results demonstrated that QS-21 showed greatest immunogenicity, reflected by robust, specific humoral response predominantly characterized IgG2a antibodies sustained cellular manifesting delayed hypersensitivity reaction. KLH- FLH-QS-21 formulations reduced tumor greater overall survival. Hemocyanins, opposed had no cytotoxic effect on either line cultured vitro, supporting idea are associated modulation immune response. Therefore, utilization would allow lower dosage achieve results.
