摘要: The purpose of this study was to develop a cost-effective approach for the determination EGFR and KRAS mutations in formalin-fixed paraffin-embedded (FFPE) non-small cell lung cancer (NSCLC) colorectal (CRC) samples from Chinese patients based on sensitive pyrosequencing (PS) technique. NSCLC CRC lines were tested determine limitation detection reproducibility PS method. In addition, 494 1099 patient assayed by evaluate or mutation patterns according clinicopathological features. assay able reproducibly detect as few 2 % mutant alleles with excellent linearity. detected 35.63 % samples, 39.76 % samples. more frequently observed be significant multivariate analysis who 65 years old younger (OR = 2.51), had nonsmoking history (OR = 3.63), adenocarcinoma (OR = 3.57), but not females (OR = 0.64). female (OR = 1.64) 50 years older (OR = 4.17), (OR = 2.41). This is first extensive validation FFPE using exons 18–21 exon 2 mutations. Our results demonstrate utility somatic clinical provide important molecular characteristics patients.
