摘要: Structural changes occurring on a millisecond time scale during uptake of DNA by cationic lipid nanocarriers are monitored time-resolved small-angle X-ray scattering (SAXS) coupled to rapid-mixing stopped-flow technique. Nanoparticles (NPs) nanochannel organization formed PEGylation, hydration, and dispersion film the fusogenic monoolein in mixture with positively charged (DOMA) PEGylated (DOPE-PEG2000) amphiphiles characterized inner cubic structure very large nanochannels favorable for upload. Ultrafast structural dynamics complexation assembly these cubosome particles neurotrophic plasmid (pDNA) is revealed thanks high brightness employed synchrotron beam. The rate constant pDNA/lipid NP estimated from dynamic roentgenograms recorded at 4 ms resolution. pDNA upload into vastly hydrated channels carriers leads fast nanoparticle-nanoparticle transition lipoplex formation involving tightly packed pDNA.
