摘要: Non-Alcoholic Fatty Liver Disease (NAFLD) is the most prevalent liver disorder worldwide, involving pathogenic mechanisms of sinusoidal endothelial cells (LSECs), hepatocytes and other cells. Here, we used a novel approach label-free Raman confocal imaging to study primary LSECs freshly isolated from livers mice with NAFLD induced by high fat diet (HFD), in comparison healthy controls. Our aim was characterize changes biochemical composition that occur single cell at subcellular level. displayed significant increase intensity marker bands nuclear DNA not associated LSEC nucleus size. A number cytoplasm were identified. However, prominent change substantial degree unsaturation LBs’ (lipid bodies) lipids NAFLD, suggesting an de novo lipogenesis unsaturated lipids. The live provided unique tool better understand disease-induced cell-specific phenotype
