摘要: Abstract We have recently studied the relative affinity of fusidate (F) for various categories albumin solutions in vitro, including plasma samples collected from hypoalbuminemic patients surgical ICU. A metabolic model has been proposed order to evaluate theoretical consequences status a patient on F kinetics. On other hand, it suggested by Peter et al. (1993) that pharmacokinetic changes observed FA with cholestasis are related an alteration metabolism free fraction (fuF), i.e. marked increase, linked subjects. In verify part our hypothesis and one (1993), we behaviour unbound administered cholestatic individuals previously studied. Data obtained vitro were compared those determined after analysis cholestasis: (1) there was significant increase C max min total between first 10th infusion; (2) AUCs fuF did not differ groups subjects; could be considered that, at equal dose, behavior is dependent upon type disease; (3) hypoalbuminemia cannot alone explain seen patients. feel results vivo product picture liver failure. Cholestasis itself does appear play preponderant role. total, comparison data this study shows limitations certain models.
