Cytotoxic Potential and Molecular Pathway Analysis of Silver Nanoparticles in Human Colon Cancer Cells HCT116

摘要: Silver nanoparticles (AgNPs) have gained attention for use in cancer therapy. In this study, AgNPs were biosynthesized using naringenin. We investigated the anti-colon activities of biogenic through transcriptome analysis RNA sequencing, and mechanisms regulating colon cell growth. The synthesized characterized UV–visible spectroscopy (UV–vis), X-ray diffraction (XRD), Fourier-transform infrared (FTIR), dynamic light scattering (DLS), transmission electron microscopy (TEM). spherical with sizes 2–10 nm. Cytotoxicity assays indicated that HCT116 colorectal cells very effective at low concentrations. viability proliferation treated 5 µg/mL reduced by 50%. Increased lactate dehydrogenase leakage (LDH), reactive oxygen species (ROS) generation, malondialdehyde (MDA), decreased dead-cell protease activity ATP generation observed. This impaired mitochondrial function DNA damage led to death. upregulated downregulated most highly ranked biological processes oxidation–reduction cell-cycle regulation, respectively. Kyoto Encyclopedia Genes Genomes (KEGG) showed GADD45G p53 pathway. Thus, AgNP tumor suppressive effects mediated apoptosis following damage, as well dysfunction arrest aberrant regulation effector proteins. It is interest mention that, best our knowledge, study first report demonstrating cellular responses molecular pathways cells.
