摘要: Cluster of differentiation (CD)133 is considered a molecular marker cancer stem cells in hepatocellular carcinoma. In the present study, effect lidamycin (LDM) on CD133 expression carcinoma (Huh7 cells) was evaluated and potential mechanism investigated. Flow cytometry analysis, as well sorting, sphere formation western-blot assays, were performed vitro to explore effects LDM expression. A subcutaneous tumor model nude mice used observe volume protein vivo. To investigate underlying mechanism, Notch signaling pathway activity detected by western blot analysis reverse transcription-quantitative polymerase chain reaction. The proportion CD133+ revealed be downregulated LDM. Sphere sorted suppressed 7 days after treatment. addition, inhibited formed from level decreased mRNA NOTCH1, Hes1 (Hes family BHLH transcription factor 1) Hey1 (Hes-related with YRPW motif genes; consequently, intracellular domain, Downregulation enhanced through combination N-[N-(3,5-difluorophenacetyl)-L-alanyl]-S-phenylglycine t-butyl ester. brief, these data suggest that suppresses via pathway, indicating benefits for further clinical application.
