摘要: A peroxidase isozyme (BP) was purified to homogeneity from broccoli stems (Brassica oleraceae var. maraton) discarded industrial processing wastes. BP specific activity 1216 ABTS [2,2′-azinobis(3-ethylbenzthiazoline-6-sulfonic acid)] units/mg, representing 466-fold that of crude extract. is a monomeric glycoprotein containing 16% carbohydrates, with molecular mass 49 kDa and an isoelectric point close 4.2. From kinetic data it showed two-substrate ping-pong mechanism, the catalytic efficiency measured as rate-limiting step free regeneration 3.4 × 106 M−1 s−1. The Km value 0.2 mM, which about 20 times lower than reported for acidic commercial horseradish (HRP). Assessment secondary structure 30% helical character, similar HRP cytochrome c peroxidase. lost only 25% after 10 min heating at 55 °C pH 6; stable in range 4 9 optimum 4.6 using substrate. ...
