摘要: Site-directed spin labeling (SDSL) in combination with electron paramagnetic resonance (EPR) spectroscopy is a well-established method that has recently grown popularity as an experimental technique, multiple applications protein and peptide science. The growth driven by development of strategies, well considerable technical advances the field, are paralleled increased availability EPR instrumentation. While requires introduction probe at well-defined position sequence, it been shown to be minimally destructive structure energetics peptide-membrane interactions. In this chapter, we describe basic approaches for using SDSL study interactions between small peptides biological membranes or membrane mimetic systems. We focus on quantify binding, topology bound peptides, characterize aggregation. Sample preparation protocols including spin-labeling methods systems also described.
