Mouse Mast Cell Protease 4 Is the Major Chymase in Murine Airways and Has a Protective Role in Allergic Airway Inflammation

摘要: It is widely established that mast cells (MCs) have a harmful role in asthma, for example by secreting various proinflammatory substances stored within their secretory granule. However, this study, we show one of the MC granule, chymase, fact has protective allergic airway inflammation, indicating MCs may possess both and activities connection with type disease. Wild-type (WT) mice lacking mouse protease 4 (mMCP-4), chymase functionally homologous to human were sensitized challenged OVA, followed assessment physiology inflammatory parameters. Our results hyperresponsiveness was significantly higher mMCP-4(-/-) as compared WT mice. Moreover, degree lung tissue inflammation markedly mMCP-4 than controls. Histological analysis revealed OVA sensitization/challenge resulted marked increased thickness smooth muscle cell (SMC) layer and, notably, SMC thickening more pronounced animals controls, thus an effect on SMCs. In support this, mMCP-4-positive located close vicinity layer, mainly upper airways, shown be major expressed these MCs.
