Allethrin toxicity on human corneal epithelial cells involves mitochondrial pathway mediated apoptosis.

摘要: Pyrethroids including allethrin are the most common commercial household insecticides. The detrimental effects caused by pyrethroids on humans gaining considerable attention. present study was aimed to elucidate of human corneal epithelial (HCE) cells. Allethrin inhibited proliferation HCE cells in a dose-dependent manner. In presence allethrin, showed membrane blebbing and nuclear fragmentation along with significant decrease mitochondrial potential resulting increased cytochrome c (Cyt c) release into cytosol. Further, flow cytometry analysis demonstrated marked increase sub G0-G1 cells, characteristic apoptosis. Increased expression pro-apoptotic protein, Bax, simultaneous anti-apoptotic Bcl-2, activation Caspase 3 evident treated addition, extracellular matrix digesting metalloproteinase 9 (MMP-9) also stimulated. Furthermore, gene inflammatory cytokines, TNF-α IL-1β observed. Taken together, these findings suggest that (IC50≈85μM) is toxic causing death through pathway.
