摘要: Immunoreactive dynorphin (ir-Dyn) and opiate-like peptides (OLP) were measured in acid (HC1) extracts of human placenta by the use an antibody to synthetic Dyn-(1-13) displacement [3H]-naloxone binding rat brain homogenates, respectively. The contained 57.6 pmoles per g ir-Dyn 134.4 naloxone equivalents. After passage extract through cartridges Sep Pak C18, half OLP was eluted with at 35% acetonitrile (ACN), rest being 60% ACN. Both fractions obtained from chromatographed separately on Sephadex G-50, ACN fraction coeluting speak that same volume as beta-endorphin. Conversely, leucine-enkephalin (Leu-Enk) these two chromatographies minimal. Dyn-immunoreactive material further purified high pressure liquid chromatography reverse phase micro-Bondapak C18 columns give three distinct peaks corresponding Dyn-(1-11), Dyn-(1-12), Our results indicate contains several forms which account for about its endogenous OLP.
