Time-dependent expression of chymase and angiotensin converting enzyme in the hamster heart under pressure overload.

摘要: The role of a dual angiotensin (Ang) II-forming pathway from the local renin system (RAS) cardiac tissue was determined in hamster model hypertrophy. Time-dependent expressions chymase and converting enzyme (ACE) genes their enzymes activities, Ang II levels were measured heart at 3 days, 4 8 weeks after pressure overload. Cardiac hypertrophy induced by an operation to constrict abdominal aorta. Compared sham-operated group, cardiomyocyte diameters hearts days overload underwent no obvious changes, while those increased markedly (p <0.01), both transcriptional ACE gradually overload, but type 1 receptor (AT1R) gene decreased. Chymase activities (U/mg) (0.441±0.040 vs. 0.175±0.014, 0.446±0.036 0.160±0.016 0.522±0.014 0.148±0.038) <0.01) (0.142±0.023 0.056±0.038, 0.317±0.017 0.079±0.016 0.466±0.010 0.098±0.003) respectively (pg/g) (98.7±4.5 71.2±4.9, 134.4±7.8 71.9±12.8 151.6±10.1 80.7±3.0) hearts, sham treatment, respectively, However, increases much higher than respective mRNA levels, also during development results suggested that increase via formation plays important hamsters caused overloaded state. Importantly, non-hypertrophied early stage (at days), could be activated mechanical stress advance its mRNA, level significantly. (Hypertens Res 2002; 25: 757-762)
