摘要: The effect of dimethylsulphoxide (DMSO) on the organization microtubular system mouse oocyte has been examined. Exposure to DMSO causes immediate appearance multiple, cold-resistant asters associated with foci pericentriolar material (PCM) normally present in oocyte. More prolonged exposure leads progressive disassembly spindle, and as a result dispersal chromosomes polar PCM occurs, tubulin polymerization becomes confined PCM-organized asters. Those astral microtubules located between cortex appear be particularly stable, resulting development lengthening radial bundles surface movement towards centre cell. In contrast, after activation generated presence remain cortical mesh. effects do not fully reversible most oocytes. We discuss implications these results both for cytoplasmic zygote, attempts at cryopreservation human oocytes therapeutic use infertility programmes.
