摘要: The process of DNA targeting or repair mutated genes within the cell, induced by specifically positioned double-strand cleavage near sequence, can be applied for gene therapy monogenic diseases. For this purpose, highly specific artificial metallonucleases are developed. They expected to important future tools modern genetics. present state art and strategies research summarized, including protein engineering ‘chemical’ nucleases. From results, we learn about basic role metal ions various ligands, binding mechanism. results collected provide useful guidance controlled enzymes use in therapy.
