摘要: Objective To evaluate the impact of monosodium urate monohydrate (MSUM) crystals on synthesis prostaglandin E2 (PGE2) by human neutrophils, and to examine some mechanisms underlying these responses. Methods The amount PGE2 released in supernatants stimulated neutrophils was evaluated enzyme immunoassay, expression cyclooxygenase 2 (COX-2) monitored immunoblot cell lysates, as well cytofluorometry buffy-coat cells. Results We observed that MSUM rapidly PGE2, with levels peaking at 1 hour. This response decreased NS-398, a specific inhibitor COX-2. We also detected constitutive COX-2 unstimulated unprimed neutrophils. rapid COX-2–dependent accumulation independent translation transcription. piceatannol, but not colchicine, blocked crystals. Conclusion These results show interaction stimulates significant mediated constitutively expressed The this study emphasize potential importance neutrophil source which may modulate, positively or negatively, inflammatory response.
