摘要: The complete nucleotide sequence of the human thymidylate synthase (TS) gene was determined. biologically active unit spans about 16 kilobase pairs (kbp) and is composed seven exons six introns. promoter region major transcriptional start sites were located within 400 base (bp) 160-180 bp, respectively, upstream from ATG initiation codon. A minigene consisting a 5'-flanking 4 kbp, coding 1.7 kpb sufficient for cell cycle-dependent expression TS when introduced into TS-deficient mutant rodent cells. does not contain "TATA," "CAAT," or consensus Sp1 binding (GC box), whereas intron 1 contains three GC boxes. G + C-rich CpG-rich extends to 1. These characteristics compared with those mouse other cycle-regulated genes.
