摘要: The trans-acting factor AP-1 is a heterodimeric complex composed of c-Jun and c-Fos family proteins which bind regulate genes containing TPA responsive enhancer element. Although binding sites have been identified within the regulatory region major histocompatibility (MHC) class I in vitro, role regulating MHC transcription vivo has not investigated previously. present study demonstrates that expression results decreased promoter activity as determined cotransfection assays an reporter construct with construct. element located between bp -440 -431 upstream initiation both functionally by direct purified c-Jun. Furthermore, over-expression reduced steady state levels endogenous RNA murine L cells approximately 10-fold. These data indicate c-Jun/AP-1 acts negative down-regulates gene expression.
