摘要: Abstract The type I cGMP-dependent protein kinase (cGK) has been shown to play a crucial role in the relaxation of vascular smooth muscle by lowering intracellular level calcium. Two isoforms cGK have described, Iα and Iβ, differing only their N-terminal parts. This report describes cloning gene PRKG1 encoding both human isoforms. is single-copy consisting 19 exons encompassing at least 220 kb. Several splice sites previously observed Drosophila melanogaster DG2 conserved PRKG1, these correlated well with boundaries between several proposed functional domains cGK. first two were encode Iα- Iβ-specific parts Using 5′-rapid amplification cDNA ends, potential for transcription initiation identified 5′ upstream exons. Northern blot analyses demonstrated distinct patterns expression Iβ different tissues.
