MALDI imaging directed laser ablation tissue microsampling for data independent acquisition proteomics.

摘要: A multimodal workflow for mass spectrometry imaging was developed that combines MALDI with protein identification and quantification by liquid chromatography tandem (LC-MS/MS). Thin tissue sections were analyzed imaging, the regions of interest (ROI) identified using a smoothing edge detection procedure. midinfrared laser at 3-μm wavelength used to remove ROI from brain section after (MALDI MSI). The captured material processed single-pot solid-phase-enhanced sample preparation (SP3) method LC-MS/MS ion mobility (IM) enhanced data independent acquisition (DIA) identify quantify proteins; more than 600 proteins identified. Using modified database included isoform post-translational modifications chain, loss initial methionine, acetylation, 14 MSI peaks Comparison Kyoto Encyclopedia Genes Genomes (KEGG) pathways achieved through an evolutionary relationships classification system.
