摘要: Abstract Dendrite cells (DCs) are a new cell type initially identified in mouse lymphoid organs. Recently, DCs have been purified from spleen. This paper demonstrates functional role of DCs: they potent stimulators the primary mixed leukocytes reaction (MLR). As few as 300-1000 doubled proliferative activity 5 X 10(6) allogeneic responder spleen cells, while 0.3-1.0 10(5) induced maximal stimulation 30- to 80-fold. Between these extremes, log MLR response increased linearly with DC numbers. dose-response assay was then used compare potency that other heterogeneous populations, many which gave curves similar slopes. The 100-300 times greater than unfractionated cells. When were fractionated by simple physical techniques, MLR-stimulating capacity subpopulations correlated closely Removal splenic B or T lymphocytes, anti-immunoglobulin anti-brain serum plus complement, did not reduce capacity. Finally, several enriched mononuclear phagocytes but lacking DCs, stimulated weakly if at all. We conclude stimulating and least 100 more effective major subclasses--i.e., lymphocytes macrophages.
