Purification of thermo and acid tolerant extracellular phytase from a new soil isolate of Amycolatopsis vancoresmycina S-12

摘要: Abstract Amycolatopsis vancoresmycina S-12, a member of soil Actinomycetes, was explored regarding its extracellular phytase. The enzyme recovered from the culture broth by ammonium sulfate fractionation and purified to near homogeneity following dialysis, DEAE Sepharose Sphadex G-100 column. 5.51 fold with final yield 14.9%. It characterized SDS PAGE, Zymogram, Native 2D PAGE as monomeric protein 53 kDa. p I  5.4. Peptide mass fingerprinting MALDI-TOF MS result indicates phytase nature. Optimum activity at 37 °C pH 5.4. stability 60 °C 8.0. is stimulated in presence Mg 2+ . plant growth promotion experiment shows significant effect, an increase 10% root 25% shoot length, resulting 2.2 biomass.
