摘要: The present study was conducted to examine the expression of tumor necrosis factor-alpha (TNF-alpha) and its receptors (types I II, designated TNFR-I TNFR-II, respectively) in human oocytes cumulus cells at mRNA protein levels. investigated using a reverse transcriptase-polymerase chain reaction (PCR)/Southern hybridization procedure. DNA-free RNA isolated from oocytes/cumulus cells, reverse-transcribed, PCR-amplified specific oligonucleotide primers based upon genomic/cDNA sequences. expected bands 303 bp 513 were observed on sequences TNF-alpha respectively, that hybridized with cDNA probes Southern blot band 368 not sequence. Similar results for level, as seen by immunoreactivity antibodies paraformaldehyde-fixed indirect immunofluorescence technique (IFT). These indicate express receptor type II (TNFR-II), (TNFR-I), both findings provide further evidence substantiate proposed physiologic role ovarian function, may lead clinical applications vitro fertilization programs diagnosis treatment infertility women, especially cases attributed dysfunction.
