摘要: Modern anatomy of the brain is primarily concerned with visualizing contrast between tissue elements and 3-dimensional (3-D) reconstruction. The first objective relies on specific neuroanatomical imaging protocols, latter proper alignment serial cross-sectional anatomy. We present a method that meets these goals at resolution allows for structural delineation in primate central nervous system. Methylene blue was administered by perfusion to intact three rhesus macaques thereby staining neuropil cortex gray nuclei situ. Subsequent dissection sectioning revealed excellent showed clear boundaries anatomical structures. captured sequential images block surface before each section cut from specimen means digital camera mounted directly over microtome stage brain. developed suite algorithms automate alignment, segmentation, normalization, intensity correction series blockface images. These were basis 3-D reconstruction brains.
