摘要: Beginning in 1988, the Chinook salmon embryo (CHSE-214) cell line was used to isolate a novel virus from spawning adult trout state of California, USA. Termed cutthroat (Oncorhynchus clarkii) (CTV), small, round not associated with disease, but subsequently found be present an increasing number populations western USA, likely by combination improved surveillance activities and shipment infected eggs new locations. Here, we report that full length genome 1988 Heenan Lake CTV consisted 7269 nucleotides positive-sense, single-stranded RNA beginning 5' untranslated region (UTR), followed three open reading frames (ORFs), 3' UTR ending polyA tail. The similar size organization Hepatitis E (HEV) which it shared highest nucleotide amino acid sequence identities. Similar genomes human, rodent or avian hepeviruses, ORF 1 encoded large, non-structural polyprotein included conserved methyltransferase, protease, helicase polymerase domains, while 2 structural capsid protein 3 phosphoprotein. Together, our data indicated clearly member family Hepeviridae, although level identity ORFs mammalian hepeviruses (13-27%) may sufficiently low warrant creation genus. We also performed phylogenetic analysis using 262nt within for 63 isolates obtained seven species reared various geographic locations While sequences fell into two genetic clades, overall diversity (less than 8.4%) many differed only 1-2 nucleotides, suggesting epidemiological link. Finally, showed able form persistently cultures CHSE-214 have use research on biology treatment hepevirus infections humans other animals.
