Facteurs de virulence de Escherichia coli adhérents et invasifs associés à la maladie de Crohn : caractérisation et régulation de leur expression

摘要: La maladie de Crohn (MC) est une affection inflammatoire chronique du tube digestif dont l'etiologie reste mal connue. Les lesions ileales patients atteints MC sont anormalement colonisees par des souches pathogenes Escherichia coli appartenant au pathovar AIEC pour Adherent-Invasive E. coli. Ces adherent et envahissent fortement les cellules epitheliales intestinales. Elles egalement capables se multiplier en macrophages d'induire secretion elevee TNF-α. L'objectif ce travail d'analyser systemes regulation la souche reference LF82 impliques dans l'expression maximale virulence sein lumiere intestinale et/ou conditions intracellulaires. Nous avons etudie co-regulation pili type 1 flagelles partant l'observation qu'un mutant n'exprimant plus perd facon concomitante sa capacite a synthetiser 1. pu montrer que cette complexe met jeu concentration intracellulaire di-GMP cyclique proteine type-histone Fis. transcriptionnelle permettent expression qui, intestinales, conferent aux un avantage rapport flore commensale. L'annotation l'analyse genome permis mettre evidence l'existence d'ilots pathogenicite, genes specifiques codants potentiels facteurs mais mutations ponctuelles, pathoadaptatives, connus permettant vraisemblablement coloniser muqueuse genetiquement predisposes developper MC.
