Transmembrane mobility of phospholipids in sickle erythrocytes: effect of deoxygenation on diffusion and asymmetry.

摘要: We studied the effect of sickling on transmembrane reorientation and distribution phospholipids in red blood cells patients homozygous for sickle cell anemia (SS). To this purpose, we followed redistribution kinetics trace amounts spin-labeled analogues natural first introduced membrane outer leaflet normal or erythrocytes exposed to air nitrogen. Deoxygenation had no lipid (AA) membranes. At atmospheric pO2, unfractionated SS were not different from cells. However, deoxygenation inducing sickling, phosphatidylcholine passive diffusion was accelerated rate adenosine triphosphate-dependent transport aminophospholipids reduced, especially phosphatidylserine. The stationary between two leaflets slightly less asymmetric, a phenomenon more pronounced with phosphatidylethanolamine. These changes rapidly reversible reoxygenation. When separated by density, both dense light exhibited properties cited above. possessed lower aminophospholipid rate. data favor relationship translocase activity phospholipid asymmetry. Sickle disease is case pathology.
