摘要: Objective: Bone marrow (BM) mesenchymal stem cells (MSCs) are being considered as potential therapeutic agents in various inflammatory autoimmune diseases for their tissue-repair and anti-inflammatory tissue-protective properties. This study investigates the reserves function, molecular proteomic profile differentiation of BM MSCs patients with active rheumatoid arthritis (RA). Methods: We evaluated frequency mononuclear cell fraction using a limiting dilution assay, proliferative/clonogenic capacity to differentiate towards osteogenic/ chondrogenic/adipogenic lineages appropriate culture conditions. also assessed characteristics terms cytokine gene protein expression, relative telomere length survival MSCs. Results: from RA (n = 26) ageand sex-matched healthy individuals 21) were similar frequency, potential, survival, immunophenotypic characteristics, profile. Patient MSCs, however, had impaired clonogenic proliferative association premature loss. Transcriptome analysis revealed differential expression genes related adhesion processes cycle progression beyond G1 phase. Previous treatment methotrexate, corticosteroids, anti-cytokine biological or other disease-modifying antiinflammatory drugs did not correlate impairment Conclusion: In spite some restrictions our findings support use autologous may have important implications ongoing efforts repair tissue injury commonly seen course disease.
