BIOPROSPECTING OF TRADITIONAL SWEET MANUFACTURING EFFLUENT FOR EXOPOLYSACHHARIDE PRODUCING BACTERIA AND THEIR BIOTECHNOLOGICAL APPLICATIONS

摘要: This work was aimed to isolate, purify, characterize and study the biological applications of extracellular polysaccharide (EPS) produced by Agrobacterium fabrum strain C 58 isolated from effluent a traditional sweet manufacturing unit. The bacterium found produce 16.21 g/L crude EPS in terms dry weight at 28 0 when brown sugar supplemented as source carbon. polysachharide further purified anion exchange chromatography on column DEAE Cellulose -52 yielding one fraction which eluted 0.2M NaCl. monosachharide composition TLC indicated it be hetero composed glucose, galactose, mannose rhamnose. FT-IR analysis proves presence biologically important functional groups alpha glycosidic linkage between individual glycosyl residues. biopolymer concentration 1 % exhibited significant lipid emulsifying capacity against various vegetable oils. effectiveness inhibiting free radicals evaluated DPPH radical scavenging appeared significant. is first report about isolation potent producers unit confirms that these samples can used potential habitat for bioprospecting polymer producing bacteria. diversity offered microorganisms diverse habitats thus renders hope screening new isolating developing polysaccharides with properties superior those existing polymers.
