Deletion of CD73 in mice leads to aortic valve dysfunction.

摘要: Abstract Aortic stenosis is known to involve inflammation and thrombosis. Changes in activity of extracellular enzyme - ecto-5′-nucleotidase (referred also as CD73) can alter inflammatory thrombotic responses. This study aimed evaluate the effect CD73 deletion mice on development aortic valve dysfunction compare it high-fat diet. Four groups (normal-diet Wild Type (WT), diet WT, normal CD73 −/−, CD73 −/−) were maintained for 15 weeks followed by echocardiographic analysis function, measurement surface activities nucleotide catabolism enzymes well alkaline phosphatase activity, mineral composition histology leaflets. CD73 −/− knock out led an increase peak flow (1.06 ± 0.26 m/s) compared WT (0.79 ± 0.26 m/s) indicating obstruction. Highest values (1.26 ± 0.31 m/s) observed mice. Histological showed morphological changes including thickening accumulation dark deposits, proved be melanin. Concentrations Ca2 +, Mg2 + PO43 − leaflets elevated Alkaline (ALP) was enhanced after ATP treatment reduced adenosine aortas incubated osteogenic medium. AMP hydrolysis below 10% WT. Activity ecto-adenosine deaminase (eADA), responsible deamination, 40% lower when Deletion leads similar that induced suggesting important role this protein maintaining heart integrity.
