摘要: Gene engineering for generating targeted mouse mutants is a key technology biomedical research. Using TALENs as sequence-specific nucleases to induce double-strand breaks, the genome can be directly modified in zygotes single step without need embryonic stem cells. By embryo microinjection of TALEN mRNAs and targeting vectors, knockout knock-in alleles generated fast efficiently. In this chapter we provide protocols application zygotes.
