Amplification of genomic DNA of arbuscular-mycorrhizal (AM) fungi by PCR using short arbitrary primers

摘要: Genomic DNA of arbuscular-mycorrhizal fungi extracted from spores was amplified with short arbitrary primers. a single-spore preparation Glomus versiforme and Gigaspora margarita provided enough material for 30 120 amplification reactions, respectively. Depending on the template-primer combination, 1–14 fragments could be separated electrophoretically. The analysis banding patterns products derived different fungal isolates defined gradient in similarity. Amplified same size were shared 9–29% species, 44–64% two species geographical regions, 60–76% close regions 77–89% lines isolate. This experimental approach, based random polymorphic (RAPD), can lead to development specific probes studying biodiversity fungi.
