摘要: Ten genomic DNA clones encoding the human leukocyte common Ag (LCA, CD45) gene were isolated by screening libraries with LCA cDNA probes. One clone contains promoter region and first two exons, as determined primer extension analyses S1 nuclease protection studies well nucleotide sequence determination. The exon does not encode a peptide, while second initiation ATG codon encodes signal peptide. other nine clones, which are separated from an unknown distance, connected span total of 73 kb. 31 exons. 33 exons encoded these 10 account for entire sequences including 5' 3' untranslated sequences. Exon 3 7 through 15 extracellular domain that to all isoforms. Differential usage 4, 5, 6, generates at least five distinct 16 transmembrane cytoplasmic antigens is composed homologous domains. Exons 17 24 domain, 25 32 domain. comparison indicated domains generated duplication several most (exon 33) carboxy terminus molecules includes sequence.
