摘要: Flow cytometry was used to study the genome size and cell cycle in different plant materials of Plantago asiatica (2n = 2C 4x) cultured vitro, including long-term callus lines. The nuclear DNA content varied from 2.97 3.45 pg/2C, depending on material analyzed. However, most cases it similar that seedling (3.3 pg), source material. This suggests vitro cultures P. produce genetically stable It seems a significantly higher or lower than leaf can be an indication lack its organogenic potential. Analysis showed there no endopolyploidization any organs; thus is likely non-polysomatic species. A high proportion nuclei (>80%; tetraploid ploidy level) maintained almost 70% calli but did not ensure their capacity. Two out 82 were octoploid they non-organogenic. In 30% endoreplication up 16C occurred. Since endopolyploid cells present as well non-organogenic calli, polysomaty level cannot considered totipotency.
