摘要: Alterations in the pattern of epidermal cell differentiation and proliferation mouse skin benign tumors were studied by two-color immunofluorescence using monospecific antibodies. Replicating cells identified 5-bromo-deoxyuridine (BrdU) pulse-labeling differentiating keratins K1 K10. In normal skin, pulse-chase experiments for 120 h revealed that replication was restricted to a single layer basal cells. did not express or K10, but these sequentially expressed post-mitotic 18 24 following DNA synthesis respectively, expressing migrated into suprabasal layers. phorbol-ester- cantharidin-stimulated hyperplastic replicating also confined compartment 1 10. papillomas induced initiation with 7,12-dimethylbenz[a]anthracene promotion 12-O-tetradecanoylphorbol-13-acetate, occurred predominantly an expanded (two four layers above basement membrane). Cells more superficial did. After pulse BrdU, differentiation-associated keratins. These other results suggest tumor escape obligatory growth arrest associated differentiation. Replication K1- K10-expressing may represent early alteration during carcinogenesis.
