摘要: Background To avoid radioisotopic labeling and permit comparison of the survival two platelet populations concurrently in one animal, we compared simultaneous recoveries times homologous rabbit platelets labeled vitro with lipophilic dyes PKH26 (red fluorescing) PKH67 (green levels biotin (low, 1 μg/ml; high, 10 μg/ml). Methods Blood samples were drawn up to 96 h postinfusion analyzed by flow cytometry. Biotin-labeled incubated phycoerythrin-streptavidin before analysis. Results Recovery PKH26-labeled at was lower (37.5%) than that PKH67-labeled (47.3%; P < 0.001). Platelet 62.4 61.9 h. Recoveries similar (86.6% 84.6%) greater those PKH-labeled (P Survival did not differ 83.3 h; 85.2 h) longer for 0.01). Labeling methods activate (measured P-selectin expression), nor they affect responses adenosine diphosphate (ADP), collagen, or thrombin. Conclusions Labeling is superior PKH dyes, useful measuring differing populations. Cytometry 47:111–117, 2002. © 2002 Wiley-Liss, Inc.
