摘要: Mycobacteriun avium ssp. paratuberculosis (MAP) is the pathogen that causes in ruminants worldwide. In Western Europe and USA it affects up to 30% of cattle herds. subclinical stage production losses occur bacterium shedded into environment. a minority animals disease ultimately progresses fatal protein loosing enteropathy. To date no vaccine available for Heat shock proteins (Hsp) are evolutionary conserved molecules have shown be immunogenic different stages bovine paratuberculosis. After natural or experimental infection immune responses Hsp70 protein, predicted protective, were predominantly cell mediated. Efficacy vaccination with MAP using DDA as an adjuvant was studied longitudinal study vaccinated experimentally infected relevant controls. Immune bacterial shedding measured almost 2 years. Single priming induced response, which clearly differed from after encounter during infection. IFN-? observed this study, however related infectious challenge rather than vaccination. A prominent prolonged antibodies restricted immunized groups. It correlated well significantly reduced bacteria immmunized challenged group, suggesting role protection. The clear divergence/discrepancy antibody enables differentiation between non-vaccinated even within herds may advantage application vaccine. use subunit vaccines implies reduction specific epitopes hence fine specificity T- B outbred animals. Th1 identified 9 epitopes, part proved promiscuous individual detected 82% (15 MHC haplotypes) MAP. Based on recognition by monoclonal epitope mapping synthetic peptides at least two linear identified, recognised calves goats well. This also provided indications present wall argued antibodies, vaccination, bind intact play direct protective effect.. course potentially protection regulatory T-cells likely role. Our studies first evidence CD4+CD25high CD4+CD25low T cells do not function (Treg) function, defined ex vivo suppressive capacity appears reside CD14+ monocytes ?? population, major populations blood contrast non-ruminant species. exact mechanism suppression Ag regulation (MAP Hsp70) has addressed future studies.
