Number of viable CD34(+) cells reinfused predicts engraftment in autologous hematopoietic stem cell transplantation.

摘要: Reduced CD34(+) cell viability due to cryopreservation has unknown effects on subsequent hematopoietic engraftment in autologous transplantation. Thirty-six consecutive peripheral stem collections were analyzed for absolute viable numbers at the time of collection and prior re-infusion. Viable cells enumerated using single platform flow cytometry molecular exclusion dye 7-amino actinomycin D. The median number was 3.6 x 10(6)/kg harvest 2.0 after thawing. When CD34(+)cells thawing 5.0 10(6)/kg, platelet 17, 12 10 days, respectively (P < 0.05 comparison group with <2.0 other two groups), neutrophil 13, 14 = NS). A minimum 10(6) cells/kg harvested 33 36 patients (92%) but only 19 (52%) met this threshold reinfusion. reduced measured re-infusion is associated may be useful monitoring loss during processing identifying risk graft failure.
