Kinetics of sulphate conjugation in extracts of human foetal liver cells in culture.

摘要: Abstract Sulphate conjugation was investigated using extracts of human foetal liver cells in culture. The three reactions which are involved sulphate were measured singly or combination: they (i) the PAPS generation catalyzed by ATP-sulphurylase and adenosine 5'-phosphosulphate (APS)-kinase, (ii) phenolsulphotransferase (PST) reaction, (iii) overall comprises above reactions. All radiometric assays employing PAP 35 S sodium sulphate. N-acetyldopamine (NADA) substrate choice although also demonstrated with dopamine 1-naphthol. Kinetic studies NADA showed two pH optima 6.7 8.6 for PST reaction while occurred maximally at 8.0. apparent K m value both same, being 38 μM, that inorganic sulphate, 107 μM 240 (measured generation, respectively) orders magnitude higher than PAPS, 2.57 μm. It possible to maintain a relatively constant level activities confluent, quiescent cultures. importance detoxification is discussed.
