摘要: A sensitive and rapid immunoassay for the detection of tumor necrosis factor (TNF) has been developed. Magnetic monodisperse polymer particles (M-280 Dynabeads) used as solid phase material, were coated with a neutralizing mouse monoclonal antibody to TNF. The Dynabeads shown have more binding capacity recombinant (r) TNF compared standard immunowells antibodies amount bound was quantified using either polyclonal or labelled 125I peroxidase. linear assay range curve form 62 4000 pg/ml, time less than 60 min. sensitivity could be increased 5-8-fold by increasing sample volume from 0.1 2 ml.
