摘要: Pituitary adenylate cyclase activating polypeptide (PACAP) isolated from ovine hypothalamus is considered to be a member of the vasoactive intestinal peptide/glucagon/secretin/growth hormone-releasing hormone family peptides. Two forms PACAP, PACAP38 and PACAP27, have been demonstrated in rat hypothalamus. The PACAP precursor contains another peptide called PACAP-related (PRP), but so far no information on this tissue exists. We developed three radioimmunoassays specific for PACAP38, PACAP27 PRP demonstrate that all preproPACAP peptides are expressed hypothalamus, PACAP38/PACAP27 ratio being around 60 PACAP38/PRP 10. HPLC analysis hypothalamic extract showed found only one form corresponding respective synthetic peptides, whereas eluted two peaks, predominant PRP1-29. cellular distribution mRNA was determined with immunohistochemistry situ hybridization histochemistry. PACAP- PRP-immunoreactive neuronal perikarya were observed medial parvocellular part paraventricular nucleus (PVN) colchicine pretreated rats. Some cell bodies magnocellular variety PVN. containing cells moderate numbers same parts nucleus. immunoreactive fibres varicosities distributed PVN periventricular These data show PVN, implying roles as regulatory
