摘要: Energy filtering transmission electron microscopy in combination with energy dispersive X-ray analysis (EDX) and quantumchemical calculations opens new possibilities for elemental bone at the ultrastructural level. The limitations of these methods, applied to botanical samples, are discussed some examples given. Ca-oxalate crystals plant cell vacuoles show a specific C K-edge loss spectrum (EELS), which allows more reliable identification than light microscopical or cytochemical methods. In dicots crystalline inclusions can be observed different compartments, identified as silicon dioxide calcium silicate by fine structure Si L2,3-edge. Their formation is on basis EEL-spectra calculations. Examples concerning heavy metal detoxification given tolerant plants.
