摘要: Apoptosis of the acinar and ductal epithelial cells salivary glands has been proposed as a mechanism possibly responsible for impairment secretory function in Sjogren’s syndrome, an organ-specific autoimmune disorder characterized by destruction these glandular structures. The presence serum autoantibodies (Abs) directed against ribonucleoproteic antigens Ro La is one classification criteria used to identify Sjogren patients, there increasing evidence direct involvement Abs tissue pathogenesis. Our recent report demonstrated that anti-Ro anti-La are able trigger extrinsic pathway apoptosis human gland cells. To better understand how exert their apoptotic effect, caspase-8 gene expression was examined primary cell (SGEC) cultures established from biopsies labial minor glands. measure mRNA changes initiating caspase-8, real-time polymerase chain reaction employed. This combined with western blot study activation detecting cleaved form poly (ADP-ribose) polymerase, downstream consequences caspases activation. Data obtained suggest determine transcriptional up-regulation caspase-8. Study SGEC experimental model may provide insight into signal transduction stimulated anti-nuclear autoantibodies.
